Organization OverviewThe Laboratory of Genome Architecture and Dynamics studies how the three-dimensional architecture of the genome reflects and regulates DNA-based processes,with a focus on short-range contacts and biophysical mechanisms of transcriptional silencing. By developing integrative models of the molecular interactions that regulate access to the genome,we hope to understand and rationally manipulate changes in chromatin architecture and gene expression during differentiation and cell reprogramming,in transitions between growth,quiescence,and senescence,and in the dysregulated cellular states found in cancer. We use mammalian primary cells and cell lines as model systems. Our approaches include sequencing-based genomic assays,biochemistry,microscopy,and modeling. We are
