DESCRIPTION
Under supervision of the principal investigator, assists generating a large-scale omics dataset of diverse bacterial species to investigate intra-species variation and strain-specific transcriptomic networks. The omics dataset includes whole-genome resequencing, transcriptome sequencing, chromatin immunoprecipitation (ChIP) sequencing, high-throughput phenotypic assay, and growth profiling. Assists handling a lab automation instrument to prepare high-throughput sequencing libraries and participate in developing an automation pipeline for NGS library generation.
Assists performing bioinformatic analysis of bacterial transcriptome, genome-wide protein-DNA interactions, and growth profiles. Assists maintaining FAIR practice of data science and accurate records of dataset and experiments. Assists performing CRISPR-mediated genome editing for mutation causality analysis. Proficiency of general lab skills in molecular biology including molecular cloning, genome editing, PCR design and optimization, microbial culture, DNA assembly, nucleic acid extraction, growth profiling and experiences in NGS library construction and bioinformatic analysis are required. Theoretical knowledge in the field of Bioengineering is required.
QUALIFICATIONS
Theoretical knowledge of molecular biology, bioengineering, cell biology.
Proficiency of performing molecular biology techniques including PCR, RT-PCR, cDNA synthesis, gene expression analysis, RNA/DNA isolation, chromatic immunoprecipitation.
Ability to perform sterile microbial cell culture techniques.
Ability to design and perform DNA cloning and assembly including restriction digest design, DNA ligation, DNA assembly.
Ability to design and perform genome editing using CRISPR/Cas9 by designing protospacers of guide-RNAs, performing mutational screening, recombineering
Ability to analyze transcriptomic data including QC, mapping, expression profiling, data normalization, DEG discovery
Ability to use computers and software to perform bioinformatic analysis including Linux, Trimmomatic, Bowtie, Samtools, BEDtools, Python with the willingness and ability to learn new software as required.
Knowledge of biostatistical analysis including hypothesis testing, significance analysis.
Detail-oriented. Ability to work with little supervision. Ability to follow verbal and/or written instructions. Ability to follow-up on previously begun procedures.
Good organizational skills and ability to prioritize work.
Enthusiasm for learning and a genuine interest in scientific research. Willingness and ability to learn new skills, research methods, and scientific principles.
Familiar in proper handling of chemical and biohazard materials. Familiarity with sterile techniques.
Ability to accurately record data and enter into appropriate medium.
Demonstrated ability to develop, follow and troubleshoot established lab protocols and lab procedures.
Ability to successfully assist with planning, organizing, prioritizing, developing, analyzing, implementing and managing research projects. Ability to set short and long-term goals for projects, and project objectives and work with others in achieving organizational goals and deadlines.
Knowledge, skill, and demonstrated experience with database design and use or willingness to learn.
Ability to read, synthesize, and communicate relevant literature in the field.
