The Diwan laboratory focuses on understanding the role of lysosomes in health and disease. Our lab has uncovered evidence for dysregulation of the lysosome machinery as a major contributor to cardiac myocyte loss in myocardial ischemia-reperfusion injury. Our work has also extended these findings to uncover evidence for lysosome dysfunction in various CNS cell types in Alzheimerâ™s disease and in pancreatic beta cells in obesity-induced diabetes, underscoring the importance of acquired lysosome dysfunction in triggering multiple chronic diseases that are predisposed by common risk factors. Using animal models, the lab is also focused on understanding how lysosome function in oocytes in moms affects cardiac risk in offspring; and whether maternal obesity increases cardiac risk by affecting lysosome function and causing epigenetic modifications in the genome.
The Diwan lab uses mice and surgical modeling tools to test hypothesis. We also use C elegans as a model system to understand the role of lysosomes in homeostasis and in stress response. Routine wet lab techniques used in the lab are molecular biology based techniques (cloning, CRISPR targeting) and biochemical (Western blotting, PCR) and histological analyses with immunohistochemistry and RNAscope based in situ hybridization.
This position works in a laboratory environment with potential exposure to biological and chemical hazards.Â The individual must be physically able to wear protective equipment and to provide standard care to research animals.
Experience working with mice, including husbandry and genotyping.
Experience with molecular biology and/or biochemical techniques.
Demonstrated ability to work independently and communicate with laboratory staff and investigators.Â
Ability to maintain complete and organized records/reports.Â
Working knowledge of computer software and general lab equipment.
