A Research Opportunity is available in the Sah Laboratory at the Washington University School of Medicine in St. Louis (Department of Internal Medicine, Cardiology) and the Center for the Investigation of Membrane Excitability Disease (CIMED) to study the mechanisms of Transient Receptor Potential (TRP) and LRRC8a/ SWELL1 ion channel regulation and signal transduction (https://sahlab.wustl.edu/). Â The laboratory uses electrophysiological (e.g. patch-clamp), imaging techniques, genetic mouse models (including CRISPR/cas9 technology), biochemical, and molecular approaches to investigate the regulation of signal transduction mechanisms of novel ion channels, including TRP/LRRC8 channels, in cardiac muscle, skeletal muscle, adipose tissue, endothelium and pancreatic beta-cells. Â Broadly speaking the overarching goal of the Sah laboratory is to understand how ion channels and mechano-signaling influence cellular and organismal metabolism. We also combine medicinal chemistry with cryo-EM structure, molecular docking, and functional studies to guide synthesis of novel ion channel modulators for the treatment of human disease.
Our laboratory models human genetic disorders in the nematode, C. elegans. The ultimate goal of our research is to better understand the disease and develop therapeutic strategies for treatment. We utilize well-established genetic tools and cutting-edge high-throughput technology to identify genetic and chemical modifiers that alter disease outcome.
Proficiency in basic molecular biology techniques, including DNA, RNA, Protein isolation and purification; PCR, reverse transcription PCR, Immunoblot, ELISA.
Experience with microscopy (dissection, compound, confocal, EM).
Working knowledge of Excel, Photoshop, and Prism.
Demonstrated ability to work independently and communicate effectively.
Ability to maintain complete and organized records.
Working knowledge of computer software and general lab equipment.
Effective verbal, written and interpersonal communication skills.
Demonstrated abilities with the Microsoft Office Suite.
Effective organizational skills and ability to prioritize workload.
Bachelor's degree in the biological sciences.
Experience with mouse handling and mouse physiology experiments.
