The Senior Research Assistant (SRA) in the Nakai lab will participate in the AAV vector and gene therapy projects conducted by the laboratory. The lab's major research focus is to elucidate the AAV vector-animal host interactions using contemporary high-throughput molecular biology technologies and develop novel AAV vectors for broader applications of this vector system to treating various human diseases.
The responsibilities of SRA include overseeing AAV vector construction and production, conducting molecular and cellular biology experiments that require advanced knowledge and skillsets and substantial wet lab benchwork experience, overseeing the lab projects and safety together with the PI and doctoral fellows, and managing the laboratory in various aspects. The management work includes inventory and ordering of lab supplies, managing and maintenance of radioisopes, training of new lab personnel and students, scheduling the lab meetings, and maintaining and updating the lab websites. These responsibilities will be assigned to SRA in addition to the daily wet lab bench work outlined below as well as, performing quality control experiments and various types of downstream analyses including Illumina sequencing. Duties will include:
Perform various molecular and cellular biology experiments: The work includes but is not limited to designing experiments together with the PI and doctoral fellows, construction and preparation of plasmid DNA, production and purification of AAV vectors, performing tissue culture cell experiments and animal experiments, extraction of DNA / RNA from samples, preparing Sanger/Illumina sequencing samples and their data analysis.
Lab Management: This includes the oversight of the daily operation of the lab including the safety monitoring and the experiments performed by lab members, troubleshooting problems lab members may come across, inventory and purchasing of lab supplies including radioisotopes, monitoring the functions of the lab equipment and the cleanliness of the lab, and the oversight of the timely washing and autoclaving of dishes, beakers and flasks, as well as the timely disposal of hazardous waste. The work also includes management of the lab website.
Collect, record, organize and verifiy the accuracy of data. Conduct literature searches and summarize information in an appropriate format for a particular study; document and interpret results of experiments and report to principal investigator. Present experimental results at lab meetings and participate in departmental and group seminars. Prepare necessary documents to help the PI write grants and scientific papers.
Bachelor's Degree with major courses in field of research and 1 to 3 years of relevant experience
Research experience in a molecular biology lab
Advanced skillsets of various molecular biology technologies including plasmid DNA cloning, DNA ligation and Gibson assembly, plasmid preparations, PCR, qPCR, DNA/RNA extraction, western blotting
Ability to work autonomously and coordinate multiple tasks simultaneously
Ability to organize a large number of samples and perform high-throughput sample processing correctly and effectively
Experience with mammalian cell culture experiments
Experience with the next-generation sequencing sample preparation
Experience with immunostaining and immunofluorescence microscopy
