A Senior Research Scientist - Basic Life position is available in the laboratory of Vittorio Sebastiano PhD, Department of Obstetrics and Gynecology at the Stanford University School of Medicine. The laboratory focuses on understanding the mechanistics of a recently developed technology called Epigenetir Reprogramming of Aging, ERA.
Aging is characterized by a gradual loss of function occurring at the molecular, cellular, tissue and organismal levels. At the chromatin level, aging associates with progressive accumulation of epigenetic errors that eventually lead to aberrant gene regulation, stem cell exhaustion, senescence, and deregulated cell/tissue homeostasis. Nuclear reprogramming to pluripotency can revert both the age and the identity of any cell to that of an embryonic cell. Recent evidence shows that transient reprogramming can ameliorate age-associated hallmarks and extend lifespan in progeroid mice. However, it is unknown how this form of rejuvenation would apply to naturally aged human cells. We have shown that transient expression of nuclear reprogramming factors, mediated by expression of mRNAs, promotes a rapid and broad amelioration of cellular aging, including resetting of epigenetic clock, reduction of the inflammatory profile in chondrocytes, and restoration of youthful regenerative response to aged, human muscle stem cells, in each case without abolishing cellular identity (Sarkar et al., 2020).
The goal of the lab is to leverage this important finding and explore potential clinical implementations of the technology and dissect the molecular mechamnism at the basis of it.
The primary role of the Senior Basic Life Research Scientist will be to supervise and guide the research activities of up to five members of the laboratory, including technicians, students and postdoctoral fellows. Responsibilities include generating and/or expanding hypotheses to further elucidate the implementation of ERA in blood, skin, or muscle cells, designing and interpretating experiments for research projects, and conducting experiments involving complex in-vitro assays, flow cytometry staining and sorting, microscopy and animal studies. In addition, the Senior Research Scientist - Basic Life is expected to review and present relevant literature. The candidate will participate in collaborative research projects with various investigators at Stanford University. The Senior Research Scientist - Basic Life will work closely with the Principal Investigator Vittorio Sebastiano, and based on prior experience and qualifications will have autonomy to make independent decisions. The candidate is required to have strong presentation and writing skills to participate in local, national and international conferences.
PhD in biological sciences preferably related to biology of skin, mucle, blood.
Minimum of five years of research laboratory experience following completion of PhD.
Proficiency with cell culture, cellular and molecular biology techniques, and animal models.
Ability to work independently and as part of a research team.
Experience in supervising or mentoring junior lab members.
Strong presentation and writing skills.
Names of 3 references should be provided.
Strong publication record in high impact journals.
Proficiency with microscopy and high-dimensional flow cytometry (8+ colors).
